Kaftrio

Kaftrio is a combination therapy containing ivacaftor, tezacaftor, and elexacaftor, developed to address the underlying cause of cystic fibrosis (CF) in individuals with specific CFTR gene variations. Instead of only easing symptoms, it works at the cellular level to improve the function of the CFTR protein, which is responsible for regulating the movement of salt and water across cell membranes.
Elexacaftor and tezacaftor help correct the processing and trafficking of the CFTR protein, enabling more of it to reach the cell surface. Ivacaftor then enhances the activity of the CFTR protein once it is in place, allowing chloride ions to pass through more effectively. Together, these actions improve hydration of mucus in organs such as the lungs and digestive system, supporting better airflow and overall organ function.
This triple-combination approach has significantly expanded treatment options for people with CF who have at least one F508del mutation, one of the most common genetic variants associated with the condition.
