Talzenna

Talzenna (talazoparib) is an oral targeted cancer therapy developed by Pfizer. It belongs to a class of drugs known as PARP inhibitors, which work by blocking the PARP (poly ADP-ribose polymerase) enzyme. This enzyme helps repair damaged DNA in cells. By inhibiting PARP, Talzenna prevents cancer cells—especially those with existing DNA repair defects—from fixing themselves, leading to cell death and slowing tumor growth.
Talzenna is primarily used in the treatment of certain types of breast cancer, particularly HER2-negative breast cancer associated with inherited BRCA gene mutations. These mutations impair normal DNA repair mechanisms, making cancer cells more vulnerable to PARP inhibition. Because of this targeted approach, Talzenna focuses on cancer cells while limiting damage to healthy cells compared to traditional chemotherapy.
Talzenna is taken orally, making it more convenient than infusion-based therapies. Its mechanism of action is considered both cytotoxic (killing cancer cells) and cytostatic (slowing their growth), providing a dual therapeutic effect. Research has shown that it can significantly delay disease progression in appropriate patient populations.
Like all cancer therapies, Talzenna can cause side effects, most commonly fatigue, anemia, nausea, and low blood cell counts. These effects occur because PARP inhibition can also impact healthy rapidly dividing cells in the body, such as those in bone marrow.
