Indications & Clinical Uses

  • Tezspire 210 mg/1.91 mL contains tezepelumab-ekko, a human monoclonal antibody that blocks thymic stromal lymphopoietin (TSLP), an upstream signal that drives airway inflammation regardless of allergy or eosinophil status.
  • It is approved as an add-on maintenance treatment for severe asthma in patients aged 12 years and older whose symptoms are not controlled by current inhaled therapies, without needing a specific blood eosinophil count or allergic trigger to qualify, and, more recently, as add-on maintenance treatment for chronic rhinosinusitis with nasal polyps that remains inadequately controlled.
  • Tezspire is co-developed and co-marketed by Amgen and AstraZeneca; it is given by subcutaneous injection once every 4 weeks, either self-injected at home after training using a prefilled syringe or pen, or given by a healthcare provider, and is not for treating a sudden asthma attack.

Side Effects

  • Tezspire has no boxed warning.
  • In asthma trials the most common effects were sore throat (pharyngitis), joint pain and back pain; in the chronic rhinosinusitis trials, sinus and upper airway infections, nosebleeds, sore throat, flu-like illness and injection-site reactions were also reported.
  • The main serious risk is hypersensitivity, including rare anaphylaxis, which can occur shortly after a dose.
  • Seek urgent medical help for swelling of the face, lips, tongue or throat, sudden difficulty breathing, widespread rash, dizziness or fainting after an injection.
  • Tezspire is not a rescue treatment - always keep your usual reliever inhaler with you for sudden symptoms.

Precautions & Warnings

  • Tezspire must not be used to treat a sudden asthma attack or status asthmaticus, and is contraindicated only in known hypersensitivity to tezepelumab or its excipients.
  • Do not stop or rapidly lower corticosteroids when starting Tezspire; any reduction should be gradual and supervised, since asthma control can worsen if steroids are withdrawn too quickly.
  • Treat any existing parasitic (helminth) infection before starting, as Tezspire may reduce the immune response needed to clear it.
  • Avoid live or live-attenuated vaccines while on treatment.
  • Data in pregnancy and breastfeeding are limited, so discuss the balance of benefit and risk individually with your prescriber.

Avoid Interactions

  • No formal drug-interaction studies have been carried out with Tezspire, and as a large monoclonal antibody it is not expected to interact through the liver enzyme pathways that affect most small-molecule medicines.
  • Commonly used asthma treatments, including inhaled corticosteroids, long-acting bronchodilators, leukotriene modifiers and theophylline, showed no clinically meaningful interaction when used alongside it in studies.
  • Avoid live or live-attenuated vaccines during treatment.
  • Tell your healthcare provider about every other medicine you take, particularly any other biologic therapy for asthma or nasal polyps, before starting Tezspire, since combining biologics is not established.
